Mobile Phase pH - 2005/11/28 18:49 I have encountered different opinions concerning buffered mobile phase pH. One side says to make the buffer solution to the required strength and pH and then add to the organic. The other side says to make the buffer solution to the required strength and add to the organic; then, you adjust the pH. Which is the proper procedure? It would appear to me that you would have to adjust the buffer first. Thanks for any input.

You may find this under another heading, but it was an oooops! I wasn't paying attention!

Mobile Phase pH - 2005/11/28 18:50 The addition of organic changes the ionization of the aqueous portion. When you mix a phosphate buffer pH 7.0 with 35% organic (Methanol 15: ACN 20) for example the effective pH is 8.8. You should always measuere pH of actual eluent.
