HPLC calibration - 2005/11/28 18:47 When is it considered acceptable to go from a multipoint calibration on a HPLC-uv system to a single point ? i have always thought it best practice to calibrate between 2 points at least- however i have been been told that a 1 point calibration is considered acceptable by the FDA for pharma. I know theoretically you should be able to go through the origin (beer-lambert)but in practice you rarely (if ever) see this and a single point calibration would surley give rise to large proportional systematic error as you approach the origin

No chemist would do a one-point calibration...

But if you want to do one, you have to be sure that you are in the linear range of the beer-lambert, that means that you have to know the extinction coefficient.
